Abstract
Protocol for sectioning OCT samples for Molecular Cartography/Resolve.
Set cryostat to -20° C. Place samples inside to acclimate to temperatures for 10 minutes.
Add OCT media to the chuck and place the sample block on top. Let freeze for 5 minutes.
Insert and fasten cryostat blade and set sectioning thickness to 30-60 um.
Place chuck in specimen station and adjust sample distance and angle to blade.
Trim sample block until desired depth is reached.
Lightly score block with a razor blade so that tissue fits within the designated scan areas on the 8-sample Resolve slides.
Section at 5 um and place section within the a scan area of the slide.
Repeat as needed to fill the slide.
Store slides at -80°C directly after sectioning.
